Novel applications of locked nucleic acids

Resumé

Locked Nucleic Acid (LNA) nucleoside triphosphates were prepared and their substrate properties for different polymerases during primer extension and PCR experiments investigated. PhusionTM High Fidelity DNA polymerase and 9°NmTM DNA polymerase readily accept LNA nucleoside 5′-triphosphates as substrates in primer extension assays. However, in PCR assays, However, in PCR assays, DNA 9oNmTM polymerase proved to be the best for amplification employing the LNA-A nucleotide.
