This project aims to unravel the genetic network involved in polarity regulation in Pseudomonas aeruginosa by a combination of synthetic lethal (Figure 2) small molecule screening (aim 1) and highdensity mutagenesis followed by deep sequencing (aim 2) of five selected polarity mutants: ΔpocA, ΔpocB, ΔtonB3, ΔflhF and ΔfimX as they are likely to be among the most important nodes in the regulatory network. These two high-throughput approaches will be used as a springboard for genetic, cell biological or biochemical analysis, depending on their outcomes. In parallel the promising results of the Caulobacter crescentus chemical genetic screening will be followed up in the context of a Masters project (aim 3).
