Epigenetics is defined as heritable traits not linked to changes in the DNA sequence. Epigenetic mechanisms organize the DNA to create a roadmap of the genome, which ensures instigation of the correct transcriptional program in a given cell type. Hereby, epigenetic regulation governs crucial processes such as proliferation, development, differentiation, stem cell renewal and genome integrity. Consequently, deregulation of epigenetic mechanisms is causally related to a number of human diseases. Whereas multiple epigenetic mechanisms have been described, the detailed knowledge of the protein complexes involved, their mode of action and regulation is relatively limited. Here, we propose a national Center of Excellence bringing together the distinct expertise of 4 laboratories in a concerted and highly ambitious effort to unveil basal mechanisms of epigenetic gene regulation fundamental to all eukaryotic organisms.
