Extrusion of xenobiotics by P-gp represents a major obstacle in relation to drug development, especially for drugs targeting cancer. Due to the fact that our current understanding of the molecular basis of the functioning of P-gp is largely unknown, blocking the functioning of this protein becomes a
very difficult task. With this research project we will obtain a fundamental molecular basic understanding of the functioning of P-gp, and further obtain atomistic insight into the binding and
modes of action of small molecules inhibiting P-gp. Based on such molecular understanding of Pglycoprotein inhibition we will devise rational design principles for small molecules specifically targeting P-gp inhibition.
